Diluent buffer for CIP phosphatase, glycerol conc? - (Jul/27/2006 )

I need ot make a Diluent buffer for CIP phosphatase and this is what it says in the recipe:
Storage Conditions:
10 mM Tris-HCl
50 mM KCl
1 mM MgCl2
0.1 mM ZnCl2
50% Glycerol
pH 8.2 @ 25°C

Do you think this 50% glycerol means to use 100% glycerol and make final concentration in the buffer 50% glycerol or to fill up to desired volume with 50% glycerol?

The diluent/storage buffer should contain 50% glycerol. It is difficult to measure 100% glycerol accurately. Use the density value of 1.261 g/mL. For a 1 mL version of the recipe, add 0.63 grams of 100% glycerol which will accurately equal 500 uL.
