RNA isolation - RNA isolation (Jul/26/2006 )

Hi, I`m isolating RNA by the Trizol method and I make a mistake, I ad ethanol 70% insted of isopropanol in the precipitating step. Can I recover my RNA?

Yes most likely. Freeze on Dry ice and/or add glycogen to aid precipitation

Yes. Add an appropriate volume of EtOH 100% in oredr to get at least a final proportion of ethanol of 75%. Add also a carrier if you consider too much dilutiong your sample for appropriate precipitation.
After that homogenize by inverting tube and keep at -80° 1h.
Spin at 4°C for 30' and then you can follow the normal procedure.
I would keep the supernatant, just in case the amount of EtOH 100% added wasn't enough (i do that because i make calculation errors sometimes and that has saved me once... rolleyes.gif )
