problem with dna fragment extraction gel - (Jul/20/2006 )

hello
i try to extract from LMA gel a DNA fragment of around 5000bp obtained by plasmid digestion
i use for this Qiaquick gel extraction kit of Qiagen, i have followed exactly the protocol of Qiagen and no recovery of DNA
I have change my solution of isopropanol (i have think it's old?), and to elute DNA i have add 30µl elution buffer and let the column stand for overnight to 4°C
but i have no recovery too
are there somebody which have same problems with extraction gel?

Hi,
I got same problem and started extracting not from one or two bands but of 6 bands or mnore bands (5mm). Yield started to increase to 15-70 ng/ul enough to do ligations.
extract as much as possible from several digestions
so u can actually see a band of the purified dna when running in an agarose gel 2 ul of ur extraction
