calculation question - (Jul/17/2006 )

goodmorning all friends
i am terribly bad in calculation so if u can please correct me in this
in my protocol for bacterial cell lysis its written to add :
1KU rLysozyme per 1 ml lysis reagent and my stock solution is : 300KU(30K U/ micron)
how much shoulkd i use for preparing 15 ml lysis buffer ??

its a basic question i know but i dont have a clue?

you want 1KU/mL, this is 1U/µL



You have 30 KU/µL


how much do you have to dilute ? 30 000 times




what's your final volume : 15 mL

then, how much do you take out of your stock solution ?






15 mL / 30000



It means you have to take 0.5µL into 15 mL

K is like in Kg or in Kcal or or or
K=Kilo=1000

Can I point out that there is no such useable unit as KU.

You mean kU. For the abbreviated form of kilo it is a lower case 'k' not an upper case 'K' which stands for Kelvin - a measurement of temperature.

Nowhere should you find kilogram written as Kg, it is kg. Same for kilodaltons (kDa), kilowatts (kW), etc. There are exceptions to avoid ambiguity. So mW is miiliwatt but MW is megawatt (not molecular weight - in this case).

but i dont get it my mentor said to add 5 micron from rLysozyme to 15 ml lysis buffer?????????????????
