troubleshooting low yield midiprep - (Jul/17/2006 )

guess the net working time for a midiprep (I use the QIAgen kit) is also around 2h, maybe a bit more...
depending on the plasmid I get ~ 285 µg of DNA from a 50 ml culture.

i dont use kit, i use alkaline lysis method and then CsCl gradient...........it takes a lot of time.......

I managed to yield about 0.8 microgram of plasmid DNA from Promega midiprep.
To obtain high yield of the DNA, you may consider to reduce the volume of the elution buffer. It is mentioned in the protocol.
