please correct me about dilution - (Jul/09/2006 )

I have to use chloramphenicol in final concentration of 50 ug/ ml in 4 litre of media.
I have a solution of 25 mg/ml of chloramphenicol.
I dissolve 250 mg in 10 ml.
from this I use 8 ml in 4 lit lb media to get a final concentation of 50 ug/ml. Is it right.

25 mg/ml = 50 ug/ml x 4000 ml


=50 ug/ml x 4000 ml/ 25 mg/ml

=50 ug /ml x 4000ml/ 25 x 1000 ug /ml.

= 200000/25000

= 8 ml.

is it right i add 8 ml of 25 mg/ml in 4 lit of culture to get 50ug/ml. I am not sure.
I am wrong, i know but where.

You want a concentration of chloramphenicol in your LB medium of 50 mikrog/ml.
So for 4 liters of LB medium this means 50 x 4000 = 200 000 mikrog = 200 mg total chloramphenicol.
To get 200 mg you just add 8 ml (= 200/25) of your stock solution (25 mg/ml) to 3992 ml of LB medium.

ur calculation and the way u calculate final concentration is absolutely right.
but i m wondering what impression make u think, that ur wrong?
donot be afraid of calculations, rather do lot of dummy calculations and get confidence.
