Unequal signal intensity across Western blot membrane - (Jul/04/2006 )

my problem is the signal on RIGHT side of membrane after taking image much stronger than on LEFT side about 5 time increasing. I use same sample, no effected and equally concentration of samples when loaded into wells.. I have repeated 4 time but they are still showing me in the same way . it look like the signal increasing from left to right side of membrane ...

anyone have Idea for solving this ....

possibly a problem with your transfer apparatus?

Could the position of the membrane during the primary and secondary's incubation made a difference.

I borrowed my friend transfer tool and test on my forth repeated ... it's still same problem ...

Yep !!... I think so about position of the membrane ... I will try to change in defferent way next run biggrin.gif

And is it impossible that the electric current is not equally.... sad.gif

May b the cover to the transfer chamber is not fitting properly and could alter current.
