Isolate splenocytes from rat - (Jun/24/2006 )

Hi,

I m try'g 2 isolate splenocytes from rat.
I m having following problems .......

1)What do u exactly mean by Spleenocytes?
2) @ what temp i should carry out dis (4 C or 26 C)?
3) what should be the ratio of Ficoll Hypaque to cell suspension?
4) I am getting mixed culture of lymphocytes, monocytes etc, How it can be selective for splenocytes?
