PH of DEPC WATER? - (Jun/19/2006 )

Does anyone know DEPC can influence PH of water? I check the PH of redistilled water either before and after DEPC treated, the PH before DEPC added is 7.11, the PH change to 3.55 after DEPC treated. is it supposed like this? or something wrong with the procedure? sad.gif sad.gif sad.gif

dont worry about that you can have Ph=8.0 with NaOH with no problem

when you're preparing MOPS solution (DEPC treated), you have to DEPCtreat, autoclave twice, and then adjust pH with NaOH. and that will be fine.

Thank you very much. Just wonder why the DEPC water should be autoclaved twice. I always just do one time autoclave for the DEPC water.

RNases are resistance to autoclave.

the purpose of autoclave isn't to destroy RNases; it is to destroy DEPC in ethanol (and an other molecule, but can't remeber the name...)
After one autoclave, some DEPC may remain in the solution, so i do twice.

Add a molar excess of RNAse A.
