This three pH system seems to work the reverse in principle as the conventional diacountinous buffer system.

the principal is the same except charges and electrodes are reversed. you can use it with proteins with a pI>5.

it should give sharp, stacked bands but, i have to say, i never ran the system. this was run by someone who i assisted when he was first running gels. he was running histone protein. he also added urea to the formula (i have that in my book, as well). this was about 33 years ago so my recollection is a little fuzzy, but he ran cylindrical gels (7cmLx5mmW, if i remember correctly) and the banding was as good as any other gel that was run at the time.
