Getting Ct values in empty wells - (Jun/14/2006 )

I am new to RT-PCR. I am getting Ct values in empty wells of the 96 well plates eventhough i am not getting Ct values in the ntc. Can you tell me why the signal is in empty well?

I'd be willing to bet that you haven't set a proper threshold. Take a look a the shape of your amplification curves. You'll see that the empty wells don't have the normal sigmoid shape of your PCR plots. Adjust the threshold so that it's above the "noise" in your empty wells, and crosses the amplification curves at a resonable location.
