ELISA with fixed bacterial cells - (Jun/13/2006 )

I have serum from inmunized mice, that I would like to analyse with ELISA. Can I use whole bacterial cells (Gram-Positive) inactivated with formaldehyde or glutaraldehyde as antigen ? will they bind to the plates, and the protein modification induced by Formaldehyde or Glutaraldehyde won't interfere with the coating process? I have seen protocols that use whole cell extracts, but since my protein of interest is expressed in the membrane, I would like to use the intact cell.

It should work. Can you UV-kill the bugs?
