qPCR Standard control - plasmid quantify (Jun/07/2006 )

I use a plasmid like a positive control during PCR smile.gif, but now I would like to use the same plasmid like standard in qPCR biggrin.gif , how can I quantify, how many plasmid copies are in 1 ul of the standard sample sad.gif .

use a spec for concentration, if you have a good prep

use molarity of your plasmid to calculate further, if ug is not close enough for you

Applied BioSystems has this great bulletin that goes through in detail the calculations for working out copy numbers.

Once you work it out you can make up a spread sheet in Exel to do all the calculations for you for different plasmids or different starting concentrations.
