Protein dosage after sucrose gradient? - (Jun/07/2006 )

Hi everyone, i need ur help
once sucrose gradient fraction are taken,
do we have to do a protein dosage to get the concentration of each fraction, and migrate the same protein quatity in western blot??
or we just take the same volume of each fraction??

it depends what you are trying to show.

if you want to show where something's distribution in the gradient then you should use equal volume.

if you want to show the relative abundance of something within the fraction then you should use equal protein load.
