Cultivate bacteriophage and isolate it's DNA from the plaque - (Jun/05/2006 )

Hello,

Does anyone know how to cultivate an phage from the bacteria E. coli K12. And how I must isolate the DNA from the plaques? Or can someone tell me, where I can find some information about the cultivation of a phage? Thanks.

Carolina Biological sells (cheaply) E. coli strains and E. coli phage stocks suitable for such experiments. They supply a manual explaining how to grow the phage. DNA is usually prepped from phage stocks by washing plaques with TE and performing a phenol/chloroform extraction.
