What’s the difference between ethanol and isopropanol precipitation? - (Jun/01/2006 )

Hi everybody!!!! biggrin.gif
What’s the difference between ethanol and isopropanol precipitation?

Does anyone knows the difference between ethanol and isopropanol precipitation of DNA?
Why some protocols recommends isopropanol and others ethanol??
I know you need less isopropanol (0.7 o 0.8 v) compared with 2 v of ethanol, but is there a more important difference???

Thanks!!

See pinned link in this forum....
EtOH vs isoprop

-karyotyper-

1. Ethanol you take double and isopropanol .7 -.8 times(this u know already) and otherwise,
2. After adding ethanol you incubate at -20 for an hour or so and after isopropanol RT for 10'!
So ISOprop looks better than etoh

-Calvin*-

calvin is right.
I would add : with IprOH you need to add more etoh 70% to wash (in theory) and pellet is slightly harder to resuspend.
