Streaks on 2D gels - (May/30/2006 )

Hi everybody.

I am running 2D gels on the proteome of Mycobacterium Tuberculosis. When I load more than 500uG of lysate I get some nasty horizontal streaks on my gel. I assume that it is the first dimension that causes the problems. I use an IPGphor unit from Amersham for the first dimension and the lysate is kept in 8M urea.

Does anybody know how to reduce to streaks?????

make sure you clarify your sample before you load it onto the first dimension. you might also want to run the first dimension longer.

What do you mean by clarify?

I will try to run the first dimension for longer time.

you can clarify by centrifugation or by filtration. you remove any solids (precipitates) from the solution.
