Is DNAse treatment essential during RNA extraction - (May/22/2006 )

I am involved in Tuberculosis diagnostics.
We are trying to detect 23s rRna of Mtb. Will somebody please clarify whether DNAse treatment is necessary prior to carrying out a RT reaction. In my opinion, the RNA being higher in copy number, the band obtained with the cDNA should be thicker than that obtained from DNA since RT+ and RT- reactions are being put up

hi
when performing RT reactions, every DNA can be a hard problem, as basically, a RNA molecule get 1 DNA molecule in counterpart. Ok there is more RNA number of molecules, but the lot of post in this forum about DNA contamination in RT reactions seems a good insight for DNAsing your RNA sample.
