Gelatin autoclaving - important (May/17/2006 )

hello guys,
i'm using gelatin to create pores on polymeric films.For this i'm autoclaving gelatin.A small doubt will it affect the structure of gelatin,i mean denatures etc.
I also mistakenly autoclaved the gelatin twice and added to teh polymer films.Will autoclaving twice affect the structure too much .ur views and comments are deeply appreciated.

I worked with gelatin in an undergraduate study and we autoclaved it without any apparent negative effects

I would say yes: Gelatin is produced from collagen, a helix protein, where the bonds are partly destroyed by heat and/or chemicals, so it is getting soluble. The more often you heat, the more the bonds for crosslinking are destroyed. Finally I guess it is not getting "hard" anymore. In the lab (and in kitchen ;-) we were advised to heat gelantin only once, other than agarose (for example to get plates or jelly...)

yep you are right.I remember 20% gelatin autoclaved once,left at room temperature for a while,got jelled.where as the the same gelatin autoclaved twice,aunder same conditions stay liquid.
But i think for the pore creation purpose it should be fine.need to look at the SEM picture for the films with autoclaved ance and twicw gelatin.Gives some ideas.
