Rna semi dry transfert overnight... - is it possible? (May/11/2006 )

Hi all.
Well i was wondering if RNA transfert from an acrylamide gel was possible overnight?...
I know that for proteins it's ok but for that purpose?... have you ever done that?

i've never done an rna electrotransfer but i wouldn't use a semi-dry overnight. the filter papers will dry up. semi-dry shouldn't be done for anything significantly greater than 1 hour (maybe 2). the semi-dry apparatus that i use (original abn, later millipore) says not to use for more than 1 hour. some of the other brands may recommend a little longer but not overnight.

For information : my boss do protein electrotransfert overnight at 10v with beautiful results.

Thanks again. That was what i though too but in the other case it would have been interesting, and my manual doesn't tell anything about that dry.gif

are his blotting papers still wet when he opens it up? maybe they dry out but after the transfer is complete.

we bought the semi-dry so that we would be able to do quick transfers. we never even thought of transfering overnight and, as i said before, the instructions said not to go over 1 hour. maybe it's because of the type of electrode abn used.

go figure.
