trypsin peptide extraction storage - (May/04/2006 )

does any body store the trypsin extractions before without drying it in the vacuum centrifuge?

i was force to store my samples in -70C before i can proceed to the drying step.

I always store the samples after(!) the speedvac. But if I´m working with the MS directly after speedvac, I stop drying at around 10µl and go on with this volume.

But in my opinion there is no problem with storring without drying, at -70C the trypsin stops digestion and there is no lose of peptides!

but will there be post modification problems or freeze-thaw will probably break the peptide chain?
will this affect the MS results?
by the way, i'll do the MALDI TOF

I have tried to store the tryptic digested peptides in -20 before speedvac, and it is ok. The longest time that i have stored the peptides was a week.
