What is the purpose of doing the RNA isolation at 4C? Does it play some role in just reducing endogenous RNAse activity? or is it actually important for layer separation?

4° is to prevent RNA degradation. But the separation of phases is better at RT when no spin.
This last point is overcome when you spin due to the fact spin accelerates phase separations.
