Dilution question - (Apr/25/2006 )

While I am making dilutions from lowest to highest,can I use the same pippet?
Somebody told me I don't need to change pippet as they are all diluted from a same sample.Is that true?If I need to change pippet from each to each,why?

If you are making a series dilution from the lowest dilution ratio (1:10) to highest dilution ratio (1:10000), then you do not need to change pipette tip.

However, if you take a aliqout of diluted sample with the lowest dilution ratio and then use the same pipette tip to take another aliqout from another sample, the bit of droplets left inside the tip may concentrated the next sample.

if you go from lowest to highest you dont have to change tips as the droplets are so small compared to the factor of ten you are increasing the dilution by, that it wont interfere with the results.

however highest to lowest, requires new tips for the opposite reason

hi,
i do the same,
while doing dilutions from lowest TO highest -- i donot change tips
moreover, if it is a low concentrated protein solution, i stick to the same tip, as we may change the concentration if the protein is sticking to each tip.
