RNA extraction without using liquid nitrogen - Can it be done? (Apr/25/2006 )

Hi,

could someone tell me if I can extract RNA, using Trizol method, without having to use liquid Nitrogen. I work with Arabidopsis, so can I just homogenize in Trizol as soon as I cut off the leaves?
Also, I am having problems resuspending my pellet, is there anything I can try?

So long as you cna disrupt the cell wall you should be able to do RNA extractions without LN2, which s just there so that you break up the cell wall.

For dissolving pellet, have you tried warming the water/ storage solution to 60C?

if you're facing pbs of pellet dissolution it may come from a too high RNA mass (so add water or your resuspending solution). Heat 56° 10 to 15' allow to redissolve.

For RNA extraction, the main pb is that the fragments should be qute very small. Homogenize with a syringe is better.
