can we add the terminaton codon in the 3' primer? - (Apr/25/2006 )

hello

i have a problem with my plasmid.i am using pRSET A as the cloning vector. i am using hind III as restriction enzyme at 3' primer. termiantion codon in the vestor is 60 bases downstream. can i add the restriction site along with 3' primer. if i dont add it will it affect the activity of the enzyme i am cloning.

thanks in advance

adding the stop codon in the 3' primer should be fine during protein expression,

although i dont know the size of the enzyme, i would say that having it as close to the natural form would be the most desirable
