DEPC before or after solution preparation?... - (Apr/21/2006 )

quite a basical question...
I want to prepare a buffer for RNA.
it's a na2hpo4 / nah2po4.
I was wonddring : can i treat the solutions with DEPC or should i depc treat the water before using the water for my buffers?....

i would prefer to prepare the buffer first and treat with DEPC.
because i will be sure of my DEPC treatment...

i've done like you describe. (in fact it's your post and in order to be sure that the vessel is also clean which decides me to do that.

in my knowledge the DEPC compoud affect only the tris buffer especially reacts with the amino groups and impeds its buffering action
