which quick ligation kit should I use ? - (Apr/20/2006 )

Hi,

I d like to switch to a quick ligation kit to save time, but I don´t know if they are really efficient for troubleshooting ligation.
Does anyone have experience with those kit ? Which one should I use ?

For sticky end ligation, I'd recommend you just skip it. Normal ligation happens quickly at room temperature. 5-30 minutes should work fine. For blunt end ligation, the kits (really the buffer with PEG) are very effective. You do need to make sure in either case that the buffer (which contains ATP) is fresh, or else add ATP explicitly.
