inclusion body had dissolved in 1M urea... - do I have to dialyse it? (Apr/18/2006 )

Agree with above. Sometimes water makes a difference, but I would be more inclined to think that it was a protein solvation problem with the urea. Try adding a little non-ionic detergent to your reduced urea buffer to allow for continued solvation effect and raise the salt levels. Your protein is generally insoluble and thats the problem.

thank you so much for the ideas! but before i try dialysis again my professor suggested I try urea/PAGE and cut out the band from the gel and then dialyze...so now im working on that but will use your idea when it comes to dialysis ..thanx a lot again! biggrin.gif
