ligation problem - (Apr/17/2006 )

hi everyone


i need some information about ligation i used control with out the insert when i do transformation

i saw so many of colony in the background i don't know what the reason

A negative control (without insert) that results in a lot of colonies when plated is either uncut vector or self ligtaion. If you digested your vector and did not purify away the uncut plasmid it will show up in your neg control. also if you have a cut vector that is blunt or has compatible sticky ends it will self ligate and again show up in your neg control.
