BAC clone amplification - (Apr/12/2006 )

Hi all

For a promoter study someone gave me a BAC clone containing the gene and promoter region, but i dont have any experience with growing up BAC clones. Can anyone help me?

Major issues include low yield (typically they are low copy origins) and fragility of the large plasmid, which can affect the recovery with some techniques, especially column or silica based techniques. Look into chloramphenicol amplification protocols to increase yield of your preps. A lot depends on what you are planning on doing with the DNA after isolation.
