Problem ratio with RNA extract - (Apr/12/2006 )

Hello guys,

I made some RNA extraction and when I mesured my OD at 260/280 I have a bad ratio (about 1,2). I realised a second precipitation and the ratio didn't change.
So anyone know what can I do to improve this ratio???

Please help me because it's so boring to see this resultttttttttt.

hi biofred ! i suspect phenol contamination in your RNA extracts. phenol in the solution changes 260/280 ratio. in your second precipitation just use chloroform and isoamyl alcohol. 24:1 ratio.

regarding phenol contamination, what is the ratio 260/230 (usually used for checking phenol contamination)?

Do not totally dry your RNA pellet, otherwise you could not get a satisfied OD at 260/280...
