homogenization for DNA extraction - (Apr/06/2006 )

Hi everybody!!
I have to homogenize big quantities of tissue (about 5 grams) to extract DNA. Final use of this DNA is PCR. Which method do you think it would be the best for that?

Why do you want to isolate lots and lots of DNA to use a tinny bit in the PCR reactions. Is there very little DNA in the tissue that you use?
