forgoten cut plasmin at room T - (Apr/05/2006 )

How bad is what i did to my minearised plasmid? I left it overnight at room T. Today i put it back into the freezer but i m scared it wont give any product after radioactive labeling using Klenow fragment. Any ideas of a worst case scenario?

you can test your plasmid quality on 0.8-1% agarose gel electroforesis

I think it should be ok but you should check it on a gel. I leave most of my DNA at room temperature and its fine. Unless there is some nuclease contamination, it shouldn't degrade.

Should be OK if you don't have DNase contamination.
I used to keep an aliquote of DNA ladder at room temperature. was convenient. It was always OK.
