Concentrating PCR product - (Mar/28/2006 )

Hi,

Is it possible to concentrate a PCR product? Cos I need to load 1 uL of 100ng of PCR product onto an applicator for the PhastSystem's gel.

Also, anyone out there who's used/ is familiar with the PhastSystem?

Yes -- I use the Qiagen PCR clean-up kit, eluting in a smaller volume. But, a PCR product is just DNA, so you can concentrate it with any method that concentrates DNA (ethanol precipitation, etc.).

There is special machinery that can do this kind of job, is called Peed abck or something like this I am not sure. It simply evaporates water so you concentrate your DNA!

It is very easy takes from 20 min to 1h (depends on the quantity of water you need to evaporate!

If you use a Speedvac, be careful. Overdrying your DNA pellet will make it a pain to resuspend.

And of course a speedvac will not remove salts or other impurities in the DNA, which may make downstream uses more difficult.

so, to be on the safe side, i'll need to run a purification process prior to speedvac, huh?
