Western Blotting band - (Mar/24/2006 )

Does anybody know what´s the mean of a very strong 50-60 KDa band in the nitrocellulose membrana after Ponceau staining?? While the other bands (smaller) are very weak.
Does it mean that my protein sample is degraded?? sad.gif

is this band across the whole blot? if so it could be an artifact caused by the reducing agent of your sample.

Hi Carol!

I have ever a strong band in this high and it's normal. If you make a beta-actin control, this whould be detected in this area to. Many Proteins overlay each other and you get a stronger band than. Thatswhy ponceau staining its not a test to control your consistent work!
