Zebrafish genomic DNA extraction - Ayuda me ! (Mar/12/2006 )

Hey everyone - new here.

I was wondering if someone might be able to give me some advice regarding extracting genomic DNA from zebrafish. Currently I am using Sigma's GenElute but getting pretty poor results. 260/280 ration too low, tried Sigma's protocol solutions, no luck.

Anyone have any experience with this ?

I recommend the good old Phenol-Chloroform method. I have tried a number of commercial high throughput DNA extraction kits. They are quick, but generally yield not so great 260/280 ratio.

BTW, which tissue are you trying to extract DNA from? fin?
