primers design for mutagenesis of two nearby sites - mutations (Mar/07/2006 )

Hi there,

I would like to make 2 mutations quite close (just separated by 3 amino acids) using Quickchange protocol and I was wondering if it is better to design a primer including the 2 mutations or making 2 differents primers containing each mutation

Any suggestions are welcome.

I never tried separated by 3 aa ... but I would try the primer including the two mutations.
I'm not telling it will work.
