Asymmetric PCR - (Mar/06/2006 )

Does any one know that why it is difficult to obtain reproducible result in asymmetric PCR each time?

The concentrations of the primers have to be perfect or else:

a) you obtain only dsDNA (too high a concentration of both primers)

cool.gif you obtain no product

thanx for ur reply.
But i kept the everything same, i mean the ration of concentration of both primers, cycle programme and the amount od DNA used. But the results were not reproducile?
