Hi.. do you actually run your HPLC with a standard before you inject your sample? Few things that you should do before you run your sample on the HPLC..
First is to attain the best detection wavelenght if your are using UV or emittence if Fluro...
Anyway.. easiest is to have a standard injection before your sample run.. Inject a known standard and you can determine the Rt of the peak and then this can be a good guide for you to know when your sample elutes... hope it is helpful...
