can anybody tell me what is the principle behind the cell lysis by freezing n thawing ???What percentage of cells can be lyzed by this process approx????

the advantage is that you can get highly concetrated lysate, eg. when looking at low abundance proteins. if i remember correctly, you can lyse whole 10cm dish in as less as 50-100ul of buffer..

Let me try this..

The principle of the cell lysis by freeze-n-thaw cycle is that the cell membrane is disrupted (i.e. cell is bursted) by the expansion of water, major component inside living cells, during freezing and cytoplasm can come out.

This is only the concept I have. If anyone know the correct answer, please correct me. Thank you.
