Real time mastermix Storage condition - The temperature for storage (Feb/14/2006 )

smile.gif Hi, I have done the real time PCR for some time and follow my colleagues' habit to store the mastermix at -20degree. But now I realize that the suggested storage temp is 2-8degree (I use the ABI Sybr-Green)mastermix. So I would like to ask if there is any problem for -20degree storage?

I only use Roche MasterMix. It's an aqueous solution, that comes freezed from manufacturer. So we store it at -20degrees. But if you would thaw it and freeze it afterwards again, growing ice crystals will destroy your enzyme. This is the reason why regular Taq-enzyme is stored in glycerol and not in aqueous solution.
After thawing your mastermix, you should store it in fridge at 8-10degrees.
