about stoichiometry analysis in Biacore expt. - (Feb/10/2006 )

Dear All,

My protein is an intact homodimer and it binds to two peptides (one peptide per monomer). Though the primary binding sites of each monomers recognize each peptides, a portion of each peptides compete to bind to the same region of the secondary binding site in the homodimer. As I want to estimate stoichiometry as well as the second binding constant of the second peptide, what kind of model I should use to determine them correctly in Biacore analysis, using Biacore3000. I am not much familiar with Biacore experiment, and this is the first time I am doing this kind of experiment. I greatly appreicate if anyone could give me the procedure/suggestion to determine these values.
