fishing out DNA - pulling out DNA with glassrod (Feb/05/2006 )

Hi all,

I extract DNA using phenol:chloro:isoamyl, after precipitating DNA in 100% ethanol, I have problem in pulling out DNA with glassrod. suggest me some possible way to fish out DNA other than centrifugation.

after you precipitate DNA add the TE buffer to that tube, let the pellet dissolve and pipete it out with a broad tip. The 100%ETOH might be giving you a very dry pellet that probably resulted in your earlier thread...

I think Pria is all over this one, but if you do decide to spool the DNA, I have found the best success with a glass pastuer pipet...you melt the tip just a little to make a shape like a croquet-hook, let it cool completely, and then you can spool the DNA around the hook

Definitely true, I am using that "method" right now, just gently swirl the DNA around the "hook" and you will be all right!

um...think twirling pasta onto your fork. if it doesn't work, then perhaps you have sheared your DNA...chromosomal DNA should be long and stringy and snotty
