About the procedure of plasmid linearization - How many positions and the proper position? (Feb/03/2006 )

Hi, all,

Whether linearized or unlinearized plasmid DNA can be used in stabel gene-transfer? Are there any different?

And also, if I want to cleave this plasmid, how many positions and which ps is much more better?

Another, if I have the bacterial culture containing plasmid now, whether the purification is the final step in the plasmid preparation.

unlinearized plazmid DNA can use for transfer of gene. ıf you wanna cleave this plasmid , you cleave one position and so gene that transfer in it stick in this position . so you have curcilar plazmid DNA again.
