HUVEC monolayer leakage testing - (Feb/01/2006 )

Dear all

We got the HUVEC from Cambrex company. We used these cells for transendothelial assay. But, unfortunately, we couldn't make sure that the endothelial monolayer was intact and had no leakage. Could any body suggest any solution for that problem?

you can measure TEER or permeability of transcellular marker

Thank you for your advice. TEER is impossible for me because we don't have the machine. But permeability assay sounds good. Do you know any kind of fluoresence linked substance that can only go through the monolayer when it has holes?

fluoresceine 300 Da, dextrans with different molecular weight conjugated to FITC or other fluorochrome, different albumins with fluorescentic tail.
the one that you will use depends on how right you want to be, i mean whether to use low or high molecular weight.
