centrifuge temperature - (Feb/01/2006 )

Does anyone know what makes the difference if we set the temperature to 4 C or RT or else during centrifugation?

It really depends on what you're spinning.

Generally, most protocols imply centrifugation at RT unless specifically stated.

While working with proteins we prefere 4C. At this temp you try to minimize degradation of your protein(s). So all our centrifuges are coled to 4C!

When for instance doing maxipreps during the isopropanol precipitation step I use the fuge @ 4°C because (as stated in the qiagen manual) this prevents overheating of the centrifuge (as this step takes 1 hour of spinning @ high speed).
