protein assay inquiry - (Jan/10/2006 )

dear all
I want to know something, during performing protein assay using bradford reagent, should the color of reagent, upon addition of the protein sample, turns into varying degrees of true blue or is it ok if it turns to a greenish blue in some samples??
it is mentioned in the manufacture suplied manual that the bradford reagent should be diluted 5 times, when I did this dilution, the low concentrations of my standarad protein samples were greenish blue. But when I performed a series of higher dilutions to the bradford reagent, i.e. 7-fold, 8-fold and 10-fold, with these dilutions I could get all my standard protein samples having different degrees of true blue depending on the concentration.
Any comments are appreciated

hi!i color depend on amount of protein in sample...i feel it s ok if it s greenish blue in sample...ur sample might contain less protein comparatively...
