I ran out of transfection agent...What to do? - (Jan/06/2006 )

Hi Guys,
I need help. I transfect MCF-7 cells with DharmaFect transfection reagent. However, today when I had finished preping for the experiment I realized that I did not have any transfection agetn. I normaly add the siRNA complex to the cells and let it sit for 72 h before changing the media and harvesting.

My cells are ready and I am repeating the same experiment fro the 3rd time. My question is:

Has anyone used lipofectamine?? That is the only thing I have on hand. If I can how can transfect. If i pull off the media after 4 hours and then let i sit for say 72 h will it be ok?? Is it worth doing it??

Since you are repeating your experiments, changing transfection reagent may render your results unproducible. So if you could not get it, just sit and wait.

Lipofectmine can certainly be used for siRNA transfection. Many of those early RNAi stuides used lipofectamine. But I have only used lipofectamine 2000. After adding transfection mixture to my cells, I don't change medium and do nothing until harvesting at 72 hrs.

thanks for the reply pcrman....I think I am just going to wait. I use lipofectamine 2000 too for my plasmid transfection...hence the question smile.gif

I just dont feel like transfection today so me shall just throw the plate off...I just want to go home and sleep but I have to stay for another 6 h unfortunately ohmy.gif

Hi

I'm doing siRNA experiment in MCF-7 and I used Lipofectamine2000 in my cell, Moreover, when I mix the siRNA-lipo complex to the cells and not chang media on cell the result is toxic for cell. So, I would like to know how your cell patant for lipofectamine or you have a good technique for experiment
