Need Help ASAP with maxiprep - Diluted plasmid with buffer ER instead of TE (qiagen kit) (Jan/04/2006 )

Hi Guys,
I need help...After 3 days of prep and plasmid isolation..by mistake I dissolved my pellet in 40 ul of ER Buffer (Qiagen kit). They say iit contains ispropanol and polyethylene glycol/phenyl ether. They do not give the percentage.

I just added another 25 ml of 70% ethanol to this and i am spinning it right now for 5 mins. The tech rep told me that the polyehtylen glycol might interefere with my OD reading. Is this true ?? and will my plasmid be ok??

May want to do several washes to remove residual PEG. No sure about the OD readings however, butI think you should be okay with recovering the DNA from the buffer usually the protocol you descibed. Let us know how it turns out.

well i did what i said i would.....just added more ethanol and spun it...my yield was ok and so were the od readings....now i have to see what happens in the cells...cross my fingers
