What? If I understand you correctly, you are treating DNA with DNAse before you sequence it? No wonder. Try not using DNAse. That's like asking about why an RNA probe won't work when one adds RNAse to the reaction after it is finished.

I have get a result from DNA sequencing that so many 'N' obtain and also oligo-'T', 'G', 'A,...But I have obtain a gel with discrete bands before sequencing, 'the extraction that run on gel have treated with DNAase, RNAase and restriction enzyme separately'.
